Inositol — particularly myo-inositol and D-chiro-inositol — has gained attention in women’s hormonal health primarily through its role in PCOS management. The connection to fibroids is indirect but real, running through the insulin-IGF-1 pathway that is directly relevant to fibroid growth.
What Inositol Does
Inositol is a naturally occurring sugar alcohol involved in insulin signalling. Specifically, myo-inositol and D-chiro-inositol act as secondary messengers in the insulin receptor pathway — supporting the cellular response to insulin and improving insulin sensitivity. Insulin resistance — where cells do not respond normally to insulin — is associated with elevated IGF-1 (insulin-like growth factor 1), which directly stimulates fibroid cell proliferation.
By improving insulin sensitivity, inositol reduces the IGF-1 burden that contributes to fibroid growth. This is the same mechanism targeted by the dietary shift from refined carbohydrates to low-glycaemic whole foods — see our fibroid diet guide — but inositol addresses it at the cellular signalling level.
Inositol and Hormonal Balance Beyond Insulin
Myo-inositol also supports FSH signalling and ovarian function — which is why it has strong evidence in PCOS for restoring regular ovulation. Regular ovulation produces adequate progesterone in the luteal phase, which counterbalances estrogen. Women with fibroids who also have irregular cycles, anovulation, or PCOS may particularly benefit from this additional hormonal support.
Evidence and Dosing
The strongest clinical evidence for inositol is in PCOS and insulin resistance contexts. Direct clinical trials in uterine fibroids are limited — the mechanism is sound, but fibroid-specific trials are not yet available. Standard dosing from PCOS literature: 2–4g myo-inositol daily (often combined with 50–200mg D-chiro-inositol in a 40:1 ratio). Inositol is generally well-tolerated; mild digestive effects are the most common side effect at higher doses.
Where Inositol Fits
Most useful for women with fibroids who also have signs of insulin resistance (difficulty losing weight, carbohydrate cravings, irregular periods), PCOS alongside fibroids, or anovulatory cycles. For these women, inositol addresses the insulin-IGF-1 pathway more directly than diet alone. See our broader natural fibroid treatment plan for how inositol fits alongside the higher-priority interventions like Vitamin D and EGCG.
